Increased Susceptibility of Cells from Cancer Patients with XV-Gonadal Dysgenesis to Simian Papovavirus 40 Transformation

چکیده
Genetically susceptible cells in culture can be transformed by oncogenic viruses. Quantitative assay methods have been developed for estimating the frequency of transformation. In an attempt to estimate the susceptibility to transformation of cells from cancer patients with XY-gonadal dysgenesis, fibroblasts from two such patients were infected with simian papovavirus 40, and transformation frequency was de termined. Results of the quantitative assay indicate that the cells from these two patients were highly susceptible to transformation. It therefore appears that XY-gonadal dysgene sis resembles in this respect the group of disorders including Fanconi's anemia, Down's syndrome, and Klinefelter's syn

Transformation frequency by simian papovavirus 40 of fibroblasts from a lung cancer patient with Klinefelter's syndrome and XY/XXY mosaicism was 3 to 10 times higher than that of fibroblasts from individuals with normal karyotypes and no history of cancer. The patient's XXY cell strain also manifested a 3-fold increase in susceptibility to transformation as compared to his XY cell strain. This ...
متن کاملThe transformation by simian virus 40 of cells from patients with mucopolysaccharidosis and from normal controls.
Fibroblasts derived from individuals with mucopolysaccharidosis, an inborn error of metabolism, have been found to be more easily transformed by simian virus 40 than are cells derived from normal individuals. The increased susceptibility does not seem to depend upon changes in glycoprotein at the cell surface. Repeated observations were necessary to demonstrate these differences, and we do not ...
متن کاملSomatic cell hybrids producing antibodies specific for the tumor antigen of simian virus 40.
We have produced somatic cell hybrids between mouse myeloma cells deficient in hypoxanthine phosphoribosyltransferase IMP: pyrophosphate phosphoribosyltransferase; EC 2.4.2.8) and spleen cells derived from mice primed with either syngeneic or allogeneic cells transformed by simian virus 40. Such hybrids produced antibodies specific for simian virus 40 tumor (T) antigen. Only four of twelve inde...
متن کاملCharacterization of tau antigens isolated from uninfected and simian virus 40-infected monkey cells and papovavirus-transformed cells.
Tau antigens (also known as cellular or nonviral tumor antigens) were detected in uninfected and simian virus 40-infected monkey cells after immunoprecipitation with serum from hamsters bearing simian virus 40-induced tumours (anti-T serum). These two proteins (56,000 daltons) were digested to similarly sized peptides with various amounts of Staphylococcus aureus V8 protease. The Tau antigen is...
